目的探讨在高频超声引导下使用Angiotech SKATER穿刺套针建立稳定的兔VX2肝癌模型的方法。方法将新西兰大白兔40只随机分为两组,一组使用开腹组织块包埋法,一组使用高频超声引导下制作兔VX2肝癌模型,观察实验兔的一般状况,并使用高频彩色多普勒观察肿瘤的生长情况及异位种植情况。结果超声引导下接种法成功率95.0%(19/20)、感染率15.0%(3/20)、异位种植率5.0%(1/20);开腹包埋法超声引导下接种法成功率100.0%(20/20)、感染率50.0%(10/20)、异位种植率5.0%(1/20)。两种方法的接种成功率、异位种植率差异均无统计学意义(P>0.05),感染率差异有统计学意义(P<0.05)。结论高频超声引导下使用Angiotech SKAT-ER穿刺套针可以建立稳定的兔VX2肝癌模型,且使用高频多普勒超声监测肿瘤模型能够更清晰的观察肿瘤的生长状况及血流情况。
Objective To investigate the method of establishing a stable rabbit VX2 hepatocellular carcinoma model using Angiotech SKATER needle guided by high-frequency ultrasound. Methods Forty New Zealand white rabbits were randomly divided into two groups. One group was treated with laparotomy. One group was made of rabbit VX2 liver cancer model guided by high frequency ultrasound. The general condition of rabbits was observed. Doppler observation of tumor growth and ectopic planting conditions. Results The success rate of inoculation under the guidance of ultrasound was 95.0% (19/20), the infection rate was 15.0% (3/20) and the rate of ectopic implantation was 5.0% (1/20). The success rate of inoculation under laparotomy 100.0% (20/20), infection rate 50.0% (10/20) and ectopic implantation rate 5.0% (1/20). There was no significant difference between the two methods in the success rate of inoculation and the rate of ectopic implantation (P> 0.05), and the difference of infection rates was statistically significant (P <0.05). Conclusion Angiotech SKAT-ER puncture trocar can establish stable rabbit VX2 liver cancer model under the guidance of high-frequency ultrasound. And the tumor growth and blood flow can be observed more clearly by using high-frequency Doppler ultrasound to monitor the tumor model.
