“人化”SCID小鼠免疫模型可望为肿瘤免疫学研究提供较理想的实验动物模型.本实验利用APAAP法,间接荧光原位杂交法及细胞毒实验观察转输入外周血淋巴细胞(PBL)在SCID小鼠体内分布及活性,比较了静脉与腹腔法转输HuPBL建立短期细胞免疫模型,现报告如下,实验方法:SPF级环境下饲养的SCID小鼠共28只,分尾静脉和腹腔转输淋巴细胞各2组,每组7只,其中2组(静脉组和腹腔组)1次转输入PBL,2周时处死,另2组再次转输淋巴细胞,4周时处死.其间,实验当日开始,每2天断尾采血,APAAP法检测淋
The immunization model of “humanized” SCID mouse is expected to provide an ideal experimental animal model for tumor immunology research.In this experiment, APAAP method, indirect fluorescence in situ hybridization and cytotoxicity assay were used to observe the effects of transfection of peripheral blood lymphocytes (PBL) The distribution and activity of SCID mice in vivo were compared, and the short-term cellular immune model was established by transplanting HuPBL by both intravenous and intraperitoneal methods.
