目的:探讨阿司匹林对巨核细胞增殖、膜抗原表达及凋亡的影响。方法:将骨髓单个核细胞接种于含不同浓度阿司匹林的H2000培养基中,其中含Tpo 50ng·ml-1、SCF 50 ng·ml-1和IL-3 10ng·ml-1,5%CO2、37℃、饱和湿度的孵育箱中培养14 d。结果:高浓度阿司匹林使巨核细胞的增殖变化明显高于低浓度,巨核细胞集落(CFU-MK)数减少,细胞丛(FFU-MK)数增加,CD41膜抗原表达率显著降低(P<0.05);AnnexinV表达率明显增高(P<0.05);DNA倍体分析的凋亡指数明显升高(P< 0.05);电镜观察可见出芽、空泡样结构和胞浆内颗粒减少。结论:高浓度阿司匹林可影响巨核细胞的正常代谢和促进其凋亡。
Objective: To investigate the effects of aspirin on megakaryocyte proliferation, membrane antigen expression and apoptosis. Methods: Bone marrow mononuclear cells were inoculated into H2000 medium containing different concentrations of aspirin, including 50ng · ml-1 of Tpo, 50 ng · ml-1 of SCF and 10ng · ml-1,5% ℃, saturated humidity incubator for 14 days. Results: The proliferation of megakaryocytes was significantly higher than that of the low concentration group. The number of megakaryocyte colonies (CFU-MK) and the number of FFU-MK cells were increased, while the expression of CD41 antigen was significantly decreased (P <0.05) ; AnnexinV expression was significantly increased (P <0.05); apoptosis index of DNA ploidy analysis was significantly increased (P <0.05); electron microscopy showed budding, vacuole-like structure and intracytoplasmic particles decreased. Conclusion: High concentration aspirin can affect the normal metabolism of megakaryocytes and promote their apoptosis.
