【目的】建立在大鼠血浆中测定毛冬青酸血药浓度的方法。【方法】大鼠尾静脉注射毛冬青酸后,依一定的时间间隔,自颈动脉采血,分离血浆,经氯仿沉淀蛋白后,以乙腈—体积分数0.2%磷酸为流动相,色谱柱为Kromasil 100-5C18(250mm×4.6mm,5μm),流速为1.0mL/min;柱温为30℃;紫外检测波长为203nm。【结果】本研究方法在2.04~51.0μg/mL范围内线性关系良好,低、中、高3个浓度的回收率为93.5%~94.8%,日内精密度sR为1.5%~4.5%、日间精密度sR为2.7%~11.1%。【结论】经过系统的方法学考察,本研究所建立的测定方法灵敏度较高、重现性良好,可用于血浆中毛冬青酸的含量测定。
【Objective】 To establish a method for the determination of the concentration of haemolitic acid in rat plasma. 【Methods】 After the tail vein injection of wool hollytic acid, the blood was collected from the carotid artery at regular intervals and the plasma was separated by precipitation with chloroform. The mobile phase consisted of acetonitrile-0.2% phosphoric acid with a volume fraction of Kromasil 100 -5 C18 (250 mm × 4.6 mm, 5 μm) at a flow rate of 1.0 mL / min. The column temperature was 30 ° C. The UV detection wavelength was 203 nm. 【Result】 The results showed good linearity in the range of 2.04-51.0 μg / mL. The recoveries of three concentrations were 93.5% -94.8% at low, medium and high concentrations. The intra-day precision sR was 1.5% -4.5% The precision sR is 2.7% ~ 11.1%. 【Conclusion】 Through the systematic methodological investigation, the assay established in this study has high sensitivity and good reproducibility, and can be used for the determination of the content of grossly active in plasma.
