目的探讨复方排毒止带汤(CDZD)对人宫颈癌SiHa细胞体外增殖的抑制作用及可能的机制。方法 15只SD大鼠随机分为三组,分别灌服CDZD煎剂(A组)、保妇康栓溶液(B组)和生理盐水(C组),7d后取得含药血清,体外与SiHa细胞培养。采用MTT法检测SiHa细胞体外增殖活性,流式细胞术检测SiHa细胞周期及细胞凋亡。结果经含药血清培养液作用72h后,A、B组SiHa细胞数量少于C组(P<0.05),A组少于B组(P<0.05)。体外培养72h,A组SiHa细胞周期停滞于S期,并诱导SiHa细胞处于凋亡的早期阶段。结论 CDZD能体外抑制SiHa细胞增殖;其机制可能与SiHa细胞周期停滞于S期和诱导细胞凋亡有关。
Objective To investigate the inhibitory effect of compound detoxification Zhidi Tang (CDZD) on the proliferation of human cervical carcinoma SiHa cells in vitro and its possible mechanism. Methods Fifteen SD rats were randomly divided into three groups and received CDZD decoction (group A), Baofukang suppository (group B) and saline (group C) Cell culture. The proliferative activity of SiHa cells in vitro was detected by MTT assay. Cell cycle and apoptosis of SiHa cells were detected by flow cytometry. Results The number of SiHa cells in groups A and B was less than that in group C (P <0.05) and group A was less than that in group B after 72 hours (P <0.05). After cultured in vitro for 72h, SiHa cell cycle in group A was arrested in S phase, and SiHa cells were induced to be in the early stage of apoptosis. Conclusion CDZD can inhibit the proliferation of SiHa cells in vitro. The mechanism may be related to the cell cycle arrest of SiHa and the induction of apoptosis.
