目的:研究肝宝胶囊对肝纤维化大鼠肝脏差异蛋白表达的影响。方法:大鼠注射二甲基亚硝胺造模,造模4周后,分为模型组和药物组,连续给药4周,处死剖取肝脏,肝脏经液氮研磨,加入裂解液,充分溶解,超声破碎处理,高速离心分离上清,提取上清液(肝脏总蛋白)。双向电泳(2-DE)分离,Image Master 2D 6.0软件进行差异蛋白表达分析,串联时间飞行质谱仪(MALDITOF/TOF)鉴定蛋白质。结果:通过对2-DE图谱蛋白斑点的匹配及对比分析,肝宝胶囊组的2-DE图谱蛋白共检出差异蛋白40个;经质谱鉴定的差异蛋白点有10个,分别为:转铁蛋白、Ⅰ型细胞骨架角蛋白10、Ⅱ型细胞骨架角蛋白1b、细胞角蛋白8多肽、甲基丙二酸半醛脱氢酶、氨甲酰磷酸合酶、热休克蛋白5、钙调素、羧酸酯酶1、谷胱甘肽S-转移酶。结论:肝宝胶囊对肝纤维化大鼠肝脏蛋白的表达与模型组比较有显著的差异;肝宝胶囊对肝纤维化大鼠肝脏蛋白的影响可能是通过多种蛋白参与的涉及多种代谢及细胞通路和细胞调控以及相互作用的复杂过程来实现的。
Objective: To study the effect of Ganbao Capsule on hepatic differential protein expression in liver fibrosis rats. Methods: Rats were injected dimethylnitrosamine into the model. After 4 weeks of modeling, the rats were divided into model group and drug group. The rats were continuously administered for 4 weeks. The liver was sacrificed and the liver was ground with liquid nitrogen. Lysate The cells were lysed, sonicated and centrifuged to isolate the supernatant (total protein of the liver). Two-dimensional electrophoresis (2-DE) separation was performed. Image Master 2D 6.0 software was used for differential protein expression analysis. Results: Through the matching and contrast analysis of 2-DE map protein spots, there were 40 differential proteins in 2-DE map protein of Ganbao capsule group and 10 differential protein spots identified by mass spectrometry, respectively Protein, type I cytoskeletal keratin 10, type II cytoskeletal keratin 1b, cytokeratin 8 polypeptide, methyl malonate semialdehyde dehydrogenase, carbamyl phosphate synthase, heat shock protein 5, calmodulin , Carboxylesterase 1, glutathione S-transferase. Conclusion: There is a significant difference between Ganbao Capsules on liver fibrosis in rats and the model group. The effect of Ganbao Capsules on hepatic protein in rats with liver fibrosis may be related to various metabolism and Cell pathways and cell regulation and the complex process of interaction to achieve.
