目的研究乙醛脱氢酶2(ALDH2)基因多态性与与颈动脉内膜中层厚度(IMT)的关系。方法从2013年4月-2014年2月体检的310例颈动脉IMT增厚者(IMT增厚组)和280例颈动脉IMT正常的健康者(对照组)外周血中提取DNA,采用DNA微阵列芯片法检测两组ALDH2基因多态性。结果 IMT增厚组与对照组基因型分布均符合Hardy-Weinberg平衡。与正常对照组相比,颈动脉IMT增厚组ALDH2基因AA/AG基因型和A等位基因频率(分别为67.4%、37.9%)明显高于对照组(分别为41.1%、22.3%),差异有统计学意义(P<0.001);logistic回归显示ALDH2基因多态性与颈动脉IMT增厚相关[OR=2.381,95%CI(1.356,4.213),P=0.004]。结论 ALDH2基因多态性与中国四川汉族人群颈动脉IMT增厚相关,ALDH2基因多态性可能在颈动脉IMT增厚遗传易感性方面具有重要作用。
Objective To investigate the association between ALDH2 gene polymorphism and carotid artery intima-media thickness (IMT). Methods DNA was extracted from peripheral blood of 310 carotid IMT thickened patients (IMT thickened group) and 280 healthy IMT control subjects (control group) from April 2013 to February 2014, and DNA microarray Array chip method was used to detect ALDH2 gene polymorphism in both groups. Results The distribution of genotypes in IMT thickening group and control group all accord with Hardy-Weinberg equilibrium. Compared with the normal control group, the frequency of AA / AG genotypes and A allele (67.4%, 37.9%, respectively) of ALDH2 gene in carotid IMT thickening group was significantly higher than that in control group (41.1% and 22.3%, respectively) The difference was statistically significant (P <0.001). Logistic regression showed that ALDH2 gene polymorphism correlated with carotid IMT thickening [OR = 2.381, 95% CI 1.356, 4.223, P = 0.004]. Conclusion ALDH2 gene polymorphism is associated with thickening of carotid artery IMT in Chinese Han population of Sichuan Province. ALDH2 gene polymorphism may play an important role in the genetic predisposition of IMT thickened carotid artery.
