目的观察甘草干姜汤加麻黄对受寒小鼠肺部AQP-1表达的影响。方法将75只SPF级昆明小鼠分为5组:常温对照组、4℃组、4℃药物组、0℃组、0℃药物组,观察实验小鼠的一般情况,7天后取实验小鼠肺部做病理切片,Western blot法检测小鼠肺部AQP-1的表达情况,并进行灰度扫描。结果与常温对照组相比,4℃组病理切片显示肺组织充血,肺泡腔内有水液潴留,肺泡隔处可见炎症细胞浸润,肺组织中AQP-1表达下降;4℃药物组病理切片显示小鼠肺部仍有充血,但肺泡腔内无渗出液,肺泡积液消失,AQP-1表达较4℃组明显增加。结论甘草干姜汤加麻黄能有效改善受寒小鼠肺部的病理损伤,受寒小鼠肺部水肿情况明显减轻,其作用机制可能是通过上调肺脏AQP-1表达,改善肺部水液代谢而发挥治疗作用。
Objective To observe the effects of Ganjiang Ginger Decoction and Ephedra on the expression of AQP-1 in the lungs of chilling mice. Methods 75 SPF Kunming mice were divided into 5 groups: normal control group, 4 ℃ group, 4 ℃ drug group, 0 ℃ group and 0 ℃ drug group. The experimental mice were observed. After 7 days, the experimental mice The lungs were made pathological sections. The expression of AQP-1 in the lungs of mice was detected by Western blot and scanned by grayscale. Results Compared with the normal control group, 4 ℃ group showed hyperemia in lung tissue, fluid retention in the alveolar space, infiltration of inflammatory cells in the alveolar septum and decreased expression of AQP-1 in lung tissue. Pathological sections at 4 ℃ Mice still have hyperemia in the lungs, but there is no exudate in the alveolar cavity, alveolar fluid disappear, AQP-1 expression was significantly increased compared with the 4 ℃ group. Conclusion Gancao Ginger Decoction and Ephedra can effectively improve the pathological injury of the lungs of chilled mice, and significantly reduce the pulmonary edema in cold mice. The mechanism may be through up-regulating the expression of AQP-1 in the lungs and improving the metabolism of lung water and fluid. Play a therapeutic role.
