目的:建立电感耦合等离子体质谱(ICP-MS)法测定透骨草中Be,Na,Mg,Al,K,Ca,V,Cr,Mn,Fe,Co,Ni,Cu,Zn,As,Se等25种无机元素的含量快速分析方法。方法:采用微波消解法处理样品,ICP-MS法,对18批次样品中25个无机元素进行测定,通过Mass Profiler Professional(MPP),MPP软件对测定结果进行主成分分析和聚类分析。结果:各元素回归方程相关系数均>0.999,加样回收率为91.1%~106.3%;不同来源、不同产地透骨草无机元素含量有较大差异,各元素随原子序数顺序呈现有规律的波浪式分布,其平均含量按由高到低的顺序前5位的依次是K(19 970.00μg·g~(-1)),Ca(15 709.00μg·g~(-1)),Mg(3 531.50μg·g~(-1)),Al(1 755.50μg·g~(-1)),Fe(1 021.00μg·g~(-1));主成分分析结果表明Be,Na,Al,Cr,Mn,Fe,Co,Zn,As,Ba,Tl,U为透骨草的特征无机元素;聚类分析将18批样品聚为2大类,广布野豌豆聚成一类;其他5种来源野豌豆聚成一类,说明广布野豌豆与其他5种野豌豆亲缘关系较远。结论:该法灵敏度高、准确性好,能同时检测多种无机元素,为中药透骨草药材的有效性、安全性评价及质量标准制订提供实验依据。
Objective: To establish an ICP-MS method for the determination of Be, Na, Mg, Al, K, Ca, V, Cr, Mn, Fe, Co, Ni, Cu, Zn, As, Se 25 kinds of inorganic elements such as rapid analysis of content. Methods: Microwave digestion method was used to treat samples and ICP-MS method to determine 25 inorganic elements in 18 batches of samples. The principal component analysis and clustering analysis were performed by using Mass Profiler Professional (MPP) and MPP software. Results: The correlation coefficients of regression equations of all elements were all> 0.999, and the recoveries were 91.1% -106.3%. The content of inorganic elements in different origins and genera of different origins were quite different. The order of the elements showed a regular wave (19 970.00 μg · g -1), Ca (15 709.00 μg · g -1), Mg (3), Mg 531.50μg · g -1, Al 1 755.50μg · g -1, Fe 1 0201μg · g -1). The results of principal component analysis showed that Be, Na, Al, Cr, Mn, Fe, Co, Zn, As, Ba, Tl and U are the characteristic inorganic elements of the genus. The cluster analysis grouped 18 batches of samples into two broad categories and broad-fielded wild peas into one category. The other five sources Wild peas converge into a category, indicating that broad-leaved wild peas and other five kinds of wild peas distant genetic relationship. Conclusion: The method has high sensitivity, good accuracy and can detect a variety of inorganic elements at the same time, providing experimental evidence for the validity, safety evaluation and quality standardization of the traditional Chinese medicine.
