目的:观察阴虚阳亢大鼠Th1/Th2细胞漂移现象、两类细胞特异性转录因子T-bet及GATA-3 mRNA地表达及左归丸对阴虚阳亢大鼠Th1/Th2细胞漂移特异性转录因子T-bet及GATA-3 mRNA的干预作用。方法:30只大鼠随机分成空白对照组(A组)、阴虚阳亢组(B组)、左归丸组(C组),B、C两组采取腺嘌呤法建立阴虚阳亢模型,然后C组用左归丸滤液灌胃,A、B两组大鼠用生理盐水灌胃,RT-PCR法分别检测用药后大鼠脾脏T-bet及GATA-3 mRNA的表达。结果:B、C组造模后、用药前T-bet mRNA表达明显上调,而GATA-3 mRNA表达下调,经左归丸组干预后,T-bet mRNA表达明显下调,而GATA-3 mRNA表达上调。结论:左归丸干预Th1/Th2细胞漂移的途径可能是通过下调Th1细胞特异性转录因子T-bet mRNA表达,上调Th2细胞特异性转录因子GATA-3 mRNA表达,使阴虚阳亢大鼠Th1/Th2细胞达到一种新的平衡。
PURPOSE: To observe the phenomenon of Th1 / Th2 cell migration, the expression of T-bet and GATA-3 mRNAs in two types of yin-deficiency and yang hyperactivity rats and the effect of Zuogui pill on the Th1 / Th2 cell migration Intervention of T-bet and GATA-3 mRNAs. Methods: Thirty rats were randomly divided into blank control group (group A), yin deficiency and hypersensitivity group (group B), Zuogui pill group (group C), B and C groups were given adenine method to establish yin deficiency and hypersensitivity model The rats in group A and B were intragastrically infused with saline solution. The expression of T-bet and GATA-3 mRNA in spleens of rats were detected by RT-PCR. Results: The expression of T-bet mRNA was significantly up-regulated and the expression of GATA-3 mRNA was down-regulated in group B and C before treatment. The expression of T-bet mRNA was down-regulated in Zuoguiwan group, while the expression of GATA-3 mRNA Increase. CONCLUSION: Zuogui pill can interfere with the Th1 / Th2 cell migration by reducing the Th1 cell-specific transcription factor T-bet mRNA expression and up-regulating the expression of Th2 cell-specific transcription factor GATA-3 mRNA, / Th2 cells to achieve a new balance.
