目的:研究活血化淤注射液Ⅰ号(HHI-Ⅰ)对肝脏缺血再灌注损伤后P38 MAP kinase的表达情况影响,为HHI-Ⅰ在临床防治肝缺血再灌注损伤的应用提供理论指导和技术支持。方法:清洁级健康雄性SD大鼠60只,体重250g左右,随机分为3组:假手术对照组(Ⅰ组)、缺血再灌注组(Ⅱ组)、HHI-Ⅰ预处理组(Ⅲ组),每组20只。分别建立大鼠肝脏缺血再灌注模型,免疫组织化学法测定肝脏缺血30min再灌注3h后组织中丝裂原活化蛋白激酶p38(P38 MAP kinase)的表达情况。结果:肝脏组织中P38 MAP kinase的表达Ⅱ、Ⅲ组高于Ⅰ组(P<0.05),Ⅲ组低于Ⅱ组(P<0.05)。结论:HHI-Ⅰ预处理可抑制P38 MAP kinase的表达,对大鼠肝脏缺血再灌注损伤有保护作用。
Objective: To investigate the effect of HHI-Ⅰ on the expression of P38 MAP kinase after hepatic ischemia-reperfusion injury and to provide theoretical guidance for the application of HHI-Ⅰ in the prevention and treatment of hepatic ischemia-reperfusion injury. Technical Support. Methods: Sixty healthy male Sprague-Dawley rats weighing 250 g were randomly divided into three groups: sham operation control group (group Ⅰ), ischemia reperfusion group (group Ⅱ), HHI-Ⅰ pretreatment group (group Ⅲ) ), Each group of 20. The model of ischemia-reperfusion in rat liver was established, and the expression of P38 MAP kinase in the tissue was detected by immunohistochemical method after 30min ischemia and reperfusion for 3h. Results: The expression of P38 MAPK in liver tissue of group Ⅱ and Ⅲ was higher than that of group Ⅰ (P <0.05), while that of group Ⅲ was lower than that of group Ⅱ (P <0.05). Conclusion: HHI-Ⅰ pretreatment can inhibit the expression of P38 MAP kinase and protect the liver from ischemia-reperfusion injury in rats.
