目的探讨熊果酸在体外对急性早幼粒白血病HL60细胞增殖抑制和诱导凋亡作用。方法采用体外细胞培养技术,荧光显微镜观察细胞结构变化、MTT法观察细胞生长抑制作用、流式细胞仪检测细胞凋亡及周期变化。结果 MTT法证实熊果酸对HL60细胞具有增殖抑制和杀伤效应,并呈浓度和时间依赖性。显微镜下可见HL60细胞出现典型凋亡细胞形态学改变。流式细胞仪检测细胞周期阻滞于G0/G1期。结论熊果酸在体外可抑制急性早幼粒白血病HL60细胞增殖,诱导细胞凋亡。
Objective To investigate the inhibitory effect of ursolic acid on the proliferation and apoptosis of acute promyelocytic leukemia HL60 cells in vitro. Methods Cell culture was used to observe the cell structure changes by fluorescence microscopy. Cell growth inhibition was observed by MTT assay. Apoptosis and cell cycle changes were detected by flow cytometry. Results MTT assay confirmed that ursolic acid had inhibitory and cytotoxic effects on HL60 cells in a concentration-and time-dependent manner. The morphological changes of typical apoptotic cells in HL60 cells were observed under the microscope. Cell cycle arrest in G0 / G1 phase was detected by flow cytometry. Conclusion Ursolic acid can inhibit the proliferation of acute promyelocytic leukemia HL60 cells in vitro and induce cell apoptosis.
