目的:证实便塞通合剂对便秘模型大鼠的通便作用是通过调节动物远端结肠AQP4表达来实现的这一假说。方法:84只SD大鼠随机分为7组:空白组、模型组、阳性对照1组(麻仁丸组)、阳性对照2组(莫沙比利组)、便塞通合剂治疗高、中、低剂量组。用复方地芬诺酯制造大鼠便秘模型,确定模型建立后,各组分别给药,每天灌胃1次,7天为1疗程,疗程间停药1天,2个疗程后动物处死,收集标本做免疫组织化学法检测。结果:AQP4在各组动物远端结肠组织均有表达,但在便塞通合剂各剂量组表达减少,尤其在中、高剂量组明显减少。便塞通高、中剂量组和莫沙比利组的AQP4阳性细胞的表达明显低于模型组和空白组(P<0.01);麻仁丸组和便塞通低剂量组大鼠AQP4的表达低于模型组大鼠(P<0.05),但与空白组比较无统计学意义(P>0.05);各给药组组间比较:便塞通高、中剂量组和莫沙比利组的AQP4阳性细胞的表达无统计学意义(P>0.05);便塞通低剂量组和麻仁丸组AQP4阳性细胞的表达无统计学意义(P>0.05),但却高于便塞通高、中剂量组和莫沙比利组的AQP4阳性细胞的表达(P<0.05)。结论:便塞通合剂对便秘模型大鼠的通便作用是通过抑制模型动物远端结肠AQP 4表达来实现的。
OBJECTIVE: To confirm that the constipation effect of Huatong Tongji in constipation model rats is achieved by regulating the expression of AQP4 in the distal colon of animals. Methods: Eighty-four SD rats were randomly divided into 7 groups: blank group, model group, positive control group 1 (Maren pill group), positive control group 2 (mosapride group) , Low dose group. Rat model of constipation was established with compound diphenoxylate. After the model was established, the rats in each group were administered with gavage once a day for 7 days for one course of treatment. The course of treatment was stopped for one day. After two courses of treatment, the animals were sacrificed and the animals were sacrificed Specimens were examined by immunohistochemistry. Results: AQP4 was expressed in the distal colon of each group of animals, but decreased in all dose groups, especially in the medium and high dose groups. The expression of AQP4 positive cells in Huitasan high dose group, middle dose group and mosapride group was significantly lower than that in model group and blank group (P <0.01) (P <0.05), but there was no significant difference between the two groups (P> 0.05) There was no significant difference in the expression of AQP4 positive cells in AQP4 positive cells (P> 0.05). There was no significant difference in the expression of AQP4 positive cells between the low dose XianQitong group and the Maren pill group (P> 0.05) Middle dose group and mosapride group AQP4 positive cells (P <0.05). CONCLUSION: The defecation effect of Huatong Tongji on constipation model rats is achieved by inhibiting the expression of AQP 4 in distal colon of model animals.
