Objective To investigate the role and significance of P53 gene in the development of mucoepidermoid carcinoma of the salivary glands. Methods The expression of P53 in 40 cases of mucoepidermoid carcinoma of the salivary glands was observed by immunohistochemistry. Results Poorly differentiated mucoepidermoid carcinoma overexpressed P53 gene, with a positive rate of 42.1%. Well-differentiated mucoepidermoid carcinoma does not express P53. The statistical analysis showed that the difference between high and poorly differentiated tumors was significant (P <0 .005). Conclusions There are differences in oncogene activation in high and poorly differentiated mucoepidermoid carcinomas. P53 gene mutation may play an important role in the occurrence of poorly differentiated mucoepidermoid carcinoma. P53 gene overexpression and mucoepidermoid carcinoma differentiation and biological behavior are closely related
