目的探讨二肾一夹肾性高血压大鼠主动脉内皮功能的变化及其机制。方法利用术后四周的肾性高血压大鼠的主动脉进行离体血管环实验,并测定主动脉cGMP的含量。结果术后四周的二肾一夹肾性高血压大鼠的主动脉cGMP含量明显减少,其离体主动脉对乙酰胆碱的舒张反应减小,而对苯肾上腺素的收缩反应显著增强。巯甲丙脯酸可增加主动脉cGMP的含量,防止这种高血压大鼠主动脉内皮功能受损。各组大鼠的离体主动脉对硝普钠的最大舒张反应无明显差别。结论提示二肾一夹肾性高血压大鼠主动脉内皮功能失调的产生主要与血管紧张素Ⅱ形成增多和一氧化氮合成、释放减少有关
Objective To investigate the changes and mechanisms of aortic endothelium function in two-kidney-one-clip hypertensive rats. Methods The isolated aortic rings of renal aorta of rats with renal hypertension were used to determine the content of cGMP in the aorta. Results The level of cGMP in the aorta of two-kidney one-clip renal hypertension rats decreased significantly after 4 weeks, and the relaxation response to acetylcholine decreased in the isolated aorta and the contractile response to phenylephrine increased significantly. Captopril increases the content of cGMP in the aorta and prevents endothelial dysfunction in the aorta of this hypertensive rat. There was no significant difference in maximal diastolic response to sodium nitroprusside between isolated aorta of rats in each group. Conclusions: The renal aortic endothelial dysfunction in two-kidney-one-clamp hypertensive rats is mainly associated with the increased formation of angiotensin Ⅱ and the decrease of nitric oxide synthesis and release
