目的 :探讨 PTEN 对人脑胶质瘤血管生成和肿瘤生长的抑制作用。 方法 :用含 PTEN基因的重组腺病毒载体Ad PTEN,体外转染人胶质瘤细胞 U87,Western印迹分析检测其表达 ,采用鸡胚绒毛尿囊膜实验检测其对血管生成的作用 ,体内实验检测腺病毒对肿瘤体积的影响 ,同时以免疫组化法检测 因子相关抗原和基质金属蛋白酶 (MMP- 2 )的表达 ,并与U87组 (不予任何处理 )和 Ad X- gal转染组相比较。结果 :U87转染 Ad PTEN腺病毒后 PTEN表达上调 ,U87组的血管指数、体内肿瘤体积、 因子相关抗原和 MMP- 2的表达病理指数均显著多于 Ad X- gal和 Ad PTEN组 (P<0 .0 5 ,P<0 .0 1 )。结论 :PTEN可抑制肿瘤血管生成 ,并抑制体内肿瘤的生长。提示 PTEN可作为有效的抗胶质瘤血管生成与肿瘤生长治疗基因
Objective: To investigate the inhibitory effect of PTEN on angiogenesis and tumor growth in human glioma. Methods: The recombinant adenovirus Ad PTEN containing PTEN gene was transfected into human glioma U87 cells in vitro and its expression was detected by Western blotting. The chick chorioallantoic membrane was used to test its effect on angiogenesis. In vivo test The effect of adenovirus on the tumor volume was examined. The expression of factor-related antigens and matrix metalloproteinase (MMP-2) was detected by immunohistochemistry. Compared with U87 group (without any treatment) and Ad-gal transfection group . Results: The expression of PTEN was up-regulated in U87 transfected with Ad PTEN adenovirus. The vascular index, in vivo tumor volume, factor-related antigen and MMP-2 expression in U87 group were significantly higher than those in Ad-gal and Ad PTEN groups (P < 0 .0 5, P <0 .0 1). Conclusion: PTEN can inhibit tumor angiogenesis and inhibit tumor growth in vivo. Tip PTEN can be used as an effective anti-glioma angiogenesis and tumor growth therapy gene
