背景与目的:Ebp1是近年发现的一种新型蛋白质,为表皮生长因子受体ErbB-3胞内近膜区结合蛋白,影响细胞增殖与分化。本研究旨在探讨人贲门癌组织中Ebp1 mRNA表达及其与临床参数的关系。方法:采用实时荧光定量PCR对120例贲门癌患者的癌组织和癌旁至少5cm处的正常贲门组织Ebp1 mRNA的表达水平进行测定。结果:73(60.83%)例贲门癌样本中Ebp1 mRNA表达下调,单因素分析显示,与贲门癌中Ebp1 mRNA的表达高度相关的临床参数包括年龄、淋巴结转移、家族史、肿瘤体积、病理分级和临床分期。多因素分析显示,年龄、家族史、肿瘤体积、病理分级及临床分期均是Ebp1表达的保护因素,而性别是Ebp1 mRNA表达的危险因素,即女性中Ebp1 mRNA表达高于男性。结论:Ebp1在贲门癌发展过程中起重要作用,可作为贲门癌病情进展的监测、转移潜能及预后评估方面的一个靶基因。
BACKGROUND & AIM: Ebp1 is a novel protein found in recent years. It is an intracellular membrane-bound protein of ErbB-3 epidermal growth factor receptor, which affects cell proliferation and differentiation. This study aimed to investigate the expression of Ebp1 mRNA in human cardiac carcinoma and its relationship with clinical parameters. Methods: The expression of Ebp1 mRNA in normal gastric cardia tissues of 120 cases of cardia cancer and normal cardia tissue of at least 5cm adjacent to cancer were measured by real-time fluorescence quantitative PCR. Results: The expression of Ebp1 mRNA was down-regulated in 73 (60.83%) cases of cardiac cancer. Univariate analysis showed that the clinical parameters highly correlated with the expression of Ebp1 mRNA in cardia cancer include age, lymph node metastasis, family history, tumor volume, Clinical stage. Multivariate analysis showed that age, family history, tumor size, pathological grade and clinical stage were the protective factors of Ebp1 expression, while sex was the risk factor of Ebp1 mRNA expression, that is, the expression of Ebp1 mRNA in women was higher than that in men. Conclusion: Ebp1 plays an important role in the development of cardia carcinoma and may be used as a target gene for the monitoring of cardiac progression, metastatic potential and prognosis.
