目的探讨系统性红斑狼疮(SLE)患者外周血单个核细胞(PBMC)中雌激素受体(ER)和热休克蛋白90(HSP90)基因的表达情况。方法分离58例SLE患者和28例正常人新鲜外周血单个核细胞,提取总RNA,利用半定量RT-PCR方法逆转录并扩增ER和HSP90,通过凝胶图像扫描系统对PCR产物的电泳条带进行扫描分析。结果 SLE患者组PBMC中ER和HSP90mRNA表达明显高于正常对照组(P均<0.05),且活动期患者的表达高于稳定期患者(P均<0.05)。结论 SLE患者中存在ER和HSP90基因的过度表达,并与疾病活动性有关。
Objective To investigate the expression of estrogen receptor (ER) and heat shock protein 90 (HSP90) in peripheral blood mononuclear cells (PBMCs) from patients with systemic lupus erythematosus (SLE). Methods 58 peripheral blood mononuclear cells from 58 SLE patients and 28 normal individuals were isolated and the total RNA was extracted. Reverse transcription and amplification of ER and HSP90 were performed by semi-quantitative RT-PCR. The electrophoresis bands of PCR products With the scan analysis. Results The expression of ER and HSP90 mRNA in PBMC of patients with SLE was significantly higher than that of the normal control group (all P <0.05), and the expression of ER and HSP90 mRNA in active patients was higher than that in stable patients (all P <0.05). Conclusion There are overexpression of ER and HSP90 genes in patients with SLE and related to disease activity.
