目的:研究淫羊藿甙(ICA)对刀豆蛋白A(ConA)刺激的小鼠T淋巴细胞体外中期和晚期活化的影响。方法:采用流式细胞术(FCM)结合双色荧光抗体染色技术分别检测中期和晚期活化标志CD25和CD71分子的表达;取中期活化的细胞上清,利用LuminexTM100分析上清中IL-2、IL-4、IL-10、TNF-α、IFN-γ的含量变化。结果:ICA在终浓度0.3、1.5、3.0μmol/L时明显抑制CD25的表达而对CD71没有明显影响;下调Th2型细胞因子及上调Th1型细胞因子的分泌量,但没有剂量依赖效应。结论:ICA下调CD25、IL-2和Th2型细胞因子水平,抑制T细胞活化,同时有一定程度的促进细胞免疫反应,因此ICA可能具有双向调节免疫平衡的作用。
Objective: To study the effect of epimedium (ICA) on the mid- and late-term activation of mouse T lymphocytes stimulated by ConA. METHODS: Flow cytometry (FCM) combined with two-color fluorescent antibody staining technique was used to detect the expression of mid- and late activation markers CD25 and CD71, respectively; middle-activated cell supernatants were taken, and IL-2 and IL-2 in the supernatant were analyzed using LuminexTM100. 4, IL-10, TNF-α, IFN-γ content changes. RESULTS: ICA inhibited the expression of CD25 at a final concentration of 0.3, 1.5, and 3.0 μmol/L, but had no significant effect on CD71. Down-regulation of Th2 cytokines and up-regulation of Th1 cytokine secretion did not have dose-dependent effects. Conclusion: ICA down-regulates the levels of CD25, IL-2 and Th2 cytokines, inhibits T cell activation, and promotes cellular immune response to a certain extent. Therefore, ICA may have a bidirectional regulation of immune balance.
