目的 :从噬菌体随机 6肽文库中筛选IL 6抗体的识别表位。方法 :利用具有IL 6中和活性的单抗C2 2 0筛选呈现于丝状噬菌体表面P3蛋白的随机 6肽文库。结果 :从噬菌体随机 6肽文库中筛选出 36株可与单抗特异结合的噬菌体克隆 ,其中多数克隆呈现核心序列SWLR ,该序列与IL 6的序列具有一定同源性。竞争结合实验和Western印迹分析 ,带有SWFNLR和HSWLRY小肽的 2株噬菌体克隆可与IL 6竞争结合单抗C2 2 0。结论 :SWFNLR和HSWLRY是IL 6单抗C2 2 0特异识别的表位。
OBJECTIVE: To screen the epitope of IL 6 antibody from phage random 6 peptide library. METHODS: A random 6-peptide library of P3 proteins on the surface of filamentous phage was screened using a monoclonal antibody C220 with IL6 neutralizing activity. RESULTS: Thirty-six phage clones that could specifically bind with McAb were screened from phage random 6-peptide library. Most of the clones showed the core sequence SWLR, which had some homology with the sequence of IL 6. Two phage clones harboring small peptides of SWFNLR and HSWLRY can compete with IL6 for binding to the monoclonal antibody C220 by competitive binding experiments and Western blot analysis. CONCLUSIONS: SWFNLR and HSWLRY are epitopes specifically recognized by IL20 monoclonal antibody C220.
