本试验是比较各种不同培养基对大蒜叶片愈伤组织的幼苗诱导的影响,找出最适宜的幼苗分化培养基。试验方法供试品种为嘉定白蒜。诱导愈伤组织的方法和培养条件见《上海农业科技》1989年第6期。幼苗诱导是将 G_4、G_5、G_7培养基中诱导出来的愈伤组织,分别移入 S_1、S_2、S_3、S_4、S_5、S_6、S_7、S_8培养基中,研究不同培养基诱导出来的愈伤组织在各种不同的培养基上发生幼苗的情况,愈伤组织年龄为45天。幼苗诱导的培养条件:培养室温度保持在20℃,以日光灯为光源,每日光照12/小时,光照强度(试管头部)为3000Lux,基础培养基是 MS 培养基,添加不同成分,组成8个培养基。
