目的建立黄芩糖蛋白的最佳提取工艺。方法从提取黄芩的pH值、温度、液料比、盐浓度、提取时间5个方面进行实验确定黄芩糖蛋白的最佳提取工艺。选取pH值、温度、液料比做正交试验,对正交试验结果进行方差分析和重复验证。采用Tris-HCl缓冲液提取法,考马斯亮蓝显色法测定黄芩糖蛋白含量,计算出糖蛋白得率。通过比较各条件下黄芩糖蛋白得率,确定黄芩糖蛋白提取的最佳条件。结果用pH=8、液料比为1:10、盐浓度0.15 mol/L的Tris-HCl缓冲液在20℃下水浴提取1.5 h,黄芩糖蛋白得率为2.94%。结论该实验首次确定黄芩糖蛋白提取最佳条件,为其糖蛋白的分离纯化与药理活性的研究提供研究基础。
Objective To establish the best extraction technology of baicalin glycoprotein. Methods The optimal extraction process of baicalin was determined experimentally from the aspects of pH value, temperature, liquid to material ratio, salt concentration and extraction time. Select the pH value, temperature, liquid ratio made orthogonal test, the orthogonal test results were analyzed by variance and repeated verification. Tris-HCl buffer extraction method, Coomassie brilliant blue colorimetric determination of baicalin protein content, calculate the glycoprotein yield. By comparing the yield of baicalin glycoprotein in each condition, the optimal conditions for baicalin glycoprotein extraction were determined. Results The yield of baicalin was 2.94% with a water bath at 20 ℃ for 1.5 h in Tris-HCl buffer solution with pH = 8, liquid-to-liquid ratio 1:10 and salt concentration 0.15 mol / L. Conclusion This experiment, for the first time, determined the optimal conditions for the extraction of baicalin glycoprotein, which provided the basis for the study on the isolation, purification and pharmacological activity of glycoprotein.
