在提取我国12个省主产棉区26个菌系gDNA的基础上,采用OPY和OPB两组随机引物,对这些菌系的基因组进行了PCR扩增和聚类分析,40个引物中,12个引物扩增出满意的适合于分析的多态性谱带。聚类分析将26个菌系分为A、B两个群,Va菌系(Verticiliumalboatrum)属于单独的A群,B群则包括了所有的大丽轮枝菌系(Verticiliumdahliae)。B群又分为Ba(Cs、Yc)及Bb两个亚群。Bb亚群下面分3个组,Bb1(AV2、AV3及Ly)、Bb2(Sa、Ji、Ke及Yc)和Bb3。Bb3是最大的一个组,包括供试的3个大丽轮枝菌落叶型的3个菌系(VD-8、T9、V44)和其他大部分非落叶型的菌系。不同的单孢之间RAPD分析的结果(引物OPY06检测出单孢之间2.06kb的差异条带)从分子水平证实了单孢间变异的发生。对河南安阳4个菌株单独聚类分析则表现出致病性测定与RAPD分析有一定的相关性。
Based on the gDNA of 26 bacterial strains in the main cotton producing areas in 12 provinces of China, the genomes of these strains were amplified by PCR and cluster analysis using random primers OPY and OPB. Of the 40 primers, 12 A primer amplified a satisfactory polymorphism band suitable for analysis. Cluster analysis divided 26 bacterial strains into A and B groups, while Va strain belonged to A group alone, while B group included all Verticilium dahliae strains. Group B is divided into Ba (Cs, Yc) and Bb two subgroups. Bb1 (AV2, AV3 and Ly), Bb2 (Sa, Ji, Ke and Yc) and Bb3 were subdivided into three subgroups. Bb3 is the largest group, consisting of 3 strains (VD-8, T9, V44) of the three deciduous Rhizoctonia species tested and the other most non-deciduous ones. The results of RAPD analysis (different bands of 2.06 kb between single spores detected by primer OPY06) among different monospores confirmed the occurrence of intermolecular mutation at the molecular level. The clustering analysis of four isolates of Anyang in Henan showed that the pathogenicity test had some correlation with RAPD analysis.
