目的探讨人乳头瘤病毒(HPV)16 E6、P21WAF1、CDK2、livin在宫颈癌中的表达及P21WAF1、CDK2、livin与E6的关系。方法采用免疫组化SP法检测HPV16 E6、P21 P21WAF1、CDK2、livin在20例宫颈癌、40例宫颈上皮内瘤变(CIN)及20例正常对照组中的表达。应用图像分析软件分别对此四个因子的表达进行平均光密度分析。结果E6、P21WAF1、CDK2、livin蛋白于宫颈癌组高于CIN1-2组及正常组(P<0.05),CIN3组高于CIN1-2组及正常组(P<0.05),宫颈癌组与CIN3组之间差异无统计学意义(P>0.05),CIN1-2组与正常组之间差异无统计学意义(P>0.05)。E6分别与P21WAF1、CDK2、livin成正相关(相关系数r分别为0.706、0.713、0.711,P<0.05)。结论E6、CDK2与宫颈癌的发生、发展密切相关,E6、CDK2异常高表达可能是宫颈疾病恶变早期事件。livin可能成为宫颈癌潜在的治疗分子靶向。
Objective To investigate the expression of human papillomavirus (HPV) 16 E6, P21WAF1, CDK2 and livin in cervical cancer and the relationship between P21WAF1, CDK2, livin and E6. Methods Immunohistochemical SP method was used to detect the expression of HPV16 E6, P21 P21WAF1, CDK2 and livin in 20 cases of cervical cancer, 40 cases of cervical intraepithelial neoplasia (CIN) and 20 cases of normal control group. The average optical density of the four factors was analyzed by image analysis software. Results The expression of E6, P21WAF1, CDK2 and livin protein in cervical cancer group was higher than that in CIN1-2 group and normal group (P <0.05), CIN3 group was higher than that in CIN1-2 group and normal group (P <0.05) There was no significant difference between groups (P> 0.05). There was no significant difference between CIN1-2 group and normal group (P> 0.05). E6 were positively correlated with P21WAF1, CDK2 and livin respectively (correlation coefficient r was 0.706,0.713,0.711, P <0.05). Conclusion E6 and CDK2 are closely related to the occurrence and development of cervical cancer. The abnormal expression of E6 and CDK2 may be the early malignant event of cervical disease. Livin may be a potential therapeutic molecule target for cervical cancer.
