目的 :观察雷公藤红素对血管平滑肌细胞 (VSMC)内游离Ca2 + 和H+ 浓度的影响。方法 :采用荧光染色法 ,以激光扫描共聚焦显微镜动态研究了不同浓度的胎牛血清对单个血管平滑肌细胞内游离Ca2 + 和H+ 浓度变化的影响效应。结果 :30 %胎牛血清可以引起细胞内一快速的、持续的游离Ca2 + 浓度升高 ,而 30 %胎牛血清加 0 3mg/L雷公藤红素则可以产生抑制细胞内游离Ca2 + 浓度升高的效应 ;2 0 %胎牛血清可以降低细胞内H+ 浓度 ,0 2mg/L雷公藤红素则可以促进细胞内H+ 浓度升高。结论 :雷公藤红素可以拮抗胎牛血清诱发的血管平滑肌细胞内游离Ca2 + 浓度升高的效应 ,其机理可能与该药物抑制了细胞膜的Ca2 + 通道有关
Objective: To observe the effects of tripterine on the concentration of free Ca2+ and H+ in vascular smooth muscle cells (VSMC). Methods : The effects of different concentrations of fetal bovine serum on the changes of free Ca2 + and H+ concentration in single vascular smooth muscle cells were studied dynamically by fluorescence staining and laser scanning confocal microscopy. RESULTS: Thirty percent fetal bovine serum could induce a rapid and sustained increase in free Ca2+ concentration in the cells, while 30 % fetal bovine serum plus 0.3 mg/L tripterine could inhibit the increase of intracellular free Ca2+ concentration. The high effect; 20% fetal bovine serum can reduce intracellular H + concentration, 0 2mg/L tripterine can promote intracellular H + concentration. Conclusion: Tripterine can antagonize the effect of fetal bovine serum on the increase of free Ca2 + concentration in vascular smooth muscle cells. The mechanism may be related to the drug’s inhibition of Ca2 + channels in cell membranes.
