在以往的实验中已经证明,CVF(中华眼镜蛇蛇毒因子)在体内具有抗血小板凝聚的作用,但在体外实验中却发现它有使血小板聚集的作用。为了探讨CVF体外引起血小板凝聚的机理,我们用放射免疫分析法测定了CVF作用后的血小板内cAMP(环磷酸腺苷)的变化。结果表明,不同剂量CVF均可使血小板内cAMP含量降低。其中生理盐水对照组血小板cAMP含量(pmol/109血小板)为25.2±6.8,CVF低浓度组为17.0±65,CVF中浓度组为16.2±7.3,CVF高浓度组为17.8±8.9。这表明CVF体外对血小板的直接作用是通过血小板内信号传导系统降低cAMP水平,导致血小板聚集。
In previous experiments, it has been shown that CVF (Chinese cobra venom factor) has the anti-platelet aggregation effect in vivo, but in vitro experiments have found that it has the effect of platelet aggregation. To investigate the mechanism of platelet aggregation caused by CVF in vitro, we measured the changes of platelet cAMP (cyclic AMP) after CVF treatment by radioimmunoassay. The results showed that different doses of CVF can reduce the platelet cAMP content. Including physiological saline control group platelet cAMP content (pmol / 109 platelet) was 25.2 ± 6.8, CVF low concentration group was 17.0 ± 65, CVF concentration group was 16.2 ± 7.3, CVF high concentration group was 17.8 ± 8.9. This suggests that the direct effect of CVF on platelets in vitro is to reduce cAMP levels via the intraplatelet signaling system resulting in platelet aggregation.
