用链尿佐菌素建立大鼠糖尿病模型,采用葡聚糖蓝-2000灌胃,作为胃肠内标志物,观察大鼠肠道传输速度,取正常对照组及模型组大鼠空肠标本,免疫组化染色观察空肠C-kit阳性Cajal间质细胞的变化,并用透射电镜观察胃窦Cajal间质细胞的超微结构改变。结果与正常组相比,糖尿病组大鼠小肠动力明显减弱(P<0.01),空肠c-kit阳性Cajal间质细胞减少(P<0.01),采用透射电镜观察显示Cajal间质细胞与其他细胞间的缝隙连接减少、结构破坏;细胞器数量明显减少,线粒体肿胀、空泡样变。结论Cajal间质细胞数量减少及超微结构变化与糖尿病肠道动力障碍有一定相关性。
The model of diabetic rats was established by streptozotocin, and gavage was performed with ganciclovir blue 2000 as the marker of gastrointestinal tract. The intestinal transit velocity of rats was observed. The jejunum specimens of normal control group and model group were obtained and immunized The changes of C-kit positive Cajal interstitial cells in jejunum were observed by histochemical staining. The ultrastructural changes of Cajal interstitial cells in gastric antrum were observed by transmission electron microscopy. Results Compared with the normal group, the small intestine motility in diabetic rats was significantly decreased (P <0.01), and the expression of c-kit positive Cajal interstitial cells in jejunum decreased (P <0.01). The transmission electron microscope showed that the interstitial cells of Cajal interstitial cells and other cells The gap connection decreased, the structure was destroyed; the number of organelles was significantly reduced, mitochondria swollen, vacuolar degeneration. Conclusions The number of interstitial cells in Cajal and ultrastructure changes have some correlation with diabetic gut dysfunction.
