大肠杆菌内毒素在体外能激活完整红细胞蛋白激酶C.为探讨内毒素在体内对细胞信息传递过程的影响以及蛋白激酶C与休克时心血管功能障碍的关系,本研究观察内毒素休克时大鼠主动脉及心肌蛋白激酶C活性的变化. 实验用Wistar大鼠,由尾静脉注射大肠杆菌内毒素(16mg/kg)建立休克模型,对照组注射等量生理盐水.注射后4h断头取主动脉及心脏,将组织匀浆,经离心及DEAE纤维素柱层析分别提取细胞浆及细胞膜蛋白激酶C,并以组蛋白(Ⅲ-S型)为底物测定酶的活性.正常大鼠主动脉组织细胞浆蛋白激酶C
