苍耳子仁粉碎后用0.15mol·L-1NaCl、0.001mol·L-1pH7.0磷酸缓冲液抽提,硫酸铵沉淀及乳糖-淀粉凝胶柱亲和层析,分离纯化得到苍耳子凝集素。经聚丙烯酰胺凝胶电泳,考马斯亮蓝R250染色,显示一条蛋白带,其溶液用苯酚-硫酸显色证明是糖蛋白。苍耳子凝集素使新鲜兔红细胞产生血凝作用所需最低浓度为6.8μg·ml-1。苍耳子凝集素的专一结合的糖为2-乙酰氨基-2-脱氧-D-半乳糖、棉子糖、乳糖和半乳糖。它对人A、B、O型血和人精子均有凝集作用。苍耳子凝集素具有较强的耐热性,反复冻融对其活性影响甚小。
The cocklebur seeds were crushed and extracted with 0.15 mol·L-1 NaCl, 0.001 mol·L-1 pH 7.0 phosphate buffer, ammonium sulfate precipitation and lactose-starch gel affinity chromatography, and the Xanthium spp. was isolated and purified. On polyacrylamide gel electrophoresis, Coomassie Brilliant Blue R250 was stained to show a protein band, and the solution was visualized with phenol-sulfuric acid to show that it was a glycoprotein. The lowest concentration of Xanthium lectin required for hemagglutination of fresh rabbit erythrocytes was 6.8 μg·ml-1. The specifically bound sugars of Xanthium lectin are 2-acetylamino-2-deoxy-D-galactose, raffinose, lactose and galactose. It has agglutination of human A, B, O blood and human sperm. Xanthium lectin has strong heat resistance, and repeated freezing and thawing has little effect on its activity.
