眼炎症是最常见的眼科疾患。环氧化酶(Cyclooxygenase)产物前列腺素(Prostaglandin, PG)是众所 周知的眼炎症介质。脂氧化酶(Lipooxygenase)代谢产物白三烯(Leukotriene, LT)是比PG更强大的炎症介质。环氧化酶1(COX1)和环氧化酶2(COX2)的发现进一步阐明了消炎痛和激素等药的药理和毒理机制。塞来昔布(Celecoxib)是第一个特异性环氧化酶-2(COX-2)抑制剂,已用于骨关节和类风湿性关节炎的症状治疗,在眼科尚未见应用报导。细胞因子除维持正常生理功能外,主要参与机体的免疫炎症及抗感染抗肿瘤等重要的病理生理过程。趋化因于是细胞因子中的一类结构和功能特殊的活性分子,被分为四个家族,即α(CXC)-、β(CC)-、γ(C)-和δ(CX3C)-趋化因子。已经证实趋化因子可使白细胞和其它细胞趋化募集于炎症或免疫反应部位,并使之活化。这些特殊细胞因子如同粘附分子和归巢受体(Homing Receptor)一样,成为新的抗炎药物研究之靶子。NO是重要的小分子免疫炎症介质。NO既参与了对病原体的杀伤防护,又参与了组织细胞的病理损伤。NO合成酶-1、2、3的发现使得该研究前进了一大步。Fas介导的凋亡过程也是机体防护组织细胞损伤破坏的机制之一。有关凋亡的增强促进剂或抑制削已经引起生物医学界的极大关注。更昔洛韦(Ganciclovir,GC
Eye inflammation is the most common eye disorder. Cyclooxygenase product Prostaglandin (PG) is a well known ocular inflammatory mediator. Lipoxygenase Metabolites Leukotriene (LT) is a more potent inflammatory mediator than PG. The discovery of cyclooxygenase 1 (COX1) and cyclooxygenase 2 (COX2) further elucidated the pharmacological and toxicological mechanisms of indomethacin and other hormones. Celecoxib is the first cyclooxygenase-2 (COX-2) inhibitor that has been used in the treatment of symptoms of osteoarthritis and rheumatoid arthritis and has not been reported in ophthalmology. In addition to maintaining normal physiological function of cytokines, the main body of the immune inflammation and anti-tumor anti-tumor and other important pathophysiological processes.
