探讨丙型肝炎病毒 ( HCV)准种在 NS2区的基因结构特征。利用逆转录 -巢式 PCR从一份 HCV慢性携带者的阳性血清及一份急性丙肝患者的血清中获得 HCV NS2全长 c DNA,将其克隆于 T载体 ,各随机挑取 5个阳性克隆进行序列测定。结果显示在克隆到的 HCV NS2全长基因中 ,慢性携带者该区序列有一个于 HCV多聚蛋白第 83 5位氨基酸的位置出现终止信号 ,丙肝患者该区序列有三个于第 83 5位氨基酸的位置出现终止信号 ,一个小于第 887位氨基酸的位置出现终止信号。在克隆到的 HCV NS2基因中存在多个 N端终止突变序列 ,提示 NS2 N端跨膜区可能与包膜蛋白区有着密切的联系
To investigate the gene structure of hepatitis C virus (HCV) quasispecies in the NS2 region.
