目的:考察归芪聪志汤对VD模型大鼠的作用及其机制。方法:采用“反复夹闭颈动脉加注射硝普钠降压再加单侧永久结扎颈总动脉法”制备VD大鼠模型,并予归芪聪志汤干预,从行为学、组织病理学、热休克蛋白(HSP)分子水平三个方面考察归芪聪志汤对VD的影响。结果:与假手术组比较,模型组大鼠定位航行潜伏期延长,空间搜索穿越平台次数减少;海马组织细胞数量较少、细胞轮廓模糊皱缩、胞浆深染、核不清晰;HSP27、HSP70mRNA及蛋白相对表达量显著升高。归芪聪志汤中、高剂量能显著改善上述指标,中剂量归芪聪志汤疗效优于脑复康。结论:归芪聪志汤能保护VD大鼠脑组织,改善其病理损伤和记忆功能,其作用机制可能与调节热休克蛋白的表达有关。
Objective: To investigate the effect of GuiQiChiZhi decoction on VD model rats and its mechanism. Methods: A rat model of VD was established by repeated occlusion of the carotid artery plus injection of sodium nitroprusside plus unilateral permanent ligation of the common carotid artery. The rats were treated with Qitongzhizhi Decoction for behavioral and histological Neo-Confucianism, heat shock protein (HSP) molecular level in three aspects of Gui Zhi clever soup on the impact of VD. Results: Compared with the sham-operation group, the rats in the model group had longer navigation latency and fewer search spaces across the platform. The number of hippocampus tissue cells was less, the cell outline was fuzzy and contracted, the cytoplasm was deeply stained and the nuclei were not clear. The HSP27, HSP70 mRNA and Protein relative expression increased significantly. Gui Qitong Zhi Tang, high-dose can significantly improve the above indicators, the dose Guiguo Zhi Zhi Tang better than the brain rehabilitation. Conclusion: Guqicongzhi decoction can protect brain tissue of VD rats and improve its pathological injury and memory function, and its mechanism may be related to the regulation of heat shock protein expression.
