目的:探讨复方大七气汤对小鼠上皮性卵巢癌的治疗效果。方法:将人卵巢癌细胞株SKOV-3注射在60只雌性BALB/C小鼠右臀部皮下建立上皮性卵巢癌模型,3周后,其中52只皮下发现包块,处死其中4只行病理组织学检查以验证其造模成功,然后将模型小鼠分为模型对照组,复方红豆杉胶囊组,复方大七气汤高剂量组、复方大七气汤中剂量组、复方大七气汤低剂量组。治疗3周末处死所有小鼠,测量癌包块体积、对癌包块进行病理切片组织学检查,同时以ELISA法检测血清糖类抗原125(CA-125)、人附睾上皮分泌蛋白4(HE4)、以SP法检测癌组织增殖细胞核抗体(PCNA)、血管内皮生长因子(VEGF)、基质金属蛋白酶-9(MMP-9)及累计光密度值。结果:复方大七气汤中剂量组卵巢癌包块体积[(18.60±0.96)mm3]、血清CA125[(6.13±0.04)U.mL-1]、HE4[(16.90±0.87)pmol.L-1]、癌组织PCNA(12.1±0.56)、VEGF(10.00±0.66)、MMP-9(14.10±0.73)和累计光密度值明显低于模型对照组与复方红豆杉胶囊治疗组(P<0.01)。结论:复方大七气汤可通过抑制细胞增殖与血管生成而对上皮性卵巢癌有治疗作用。
Objective: To investigate the therapeutic effect of Fufangqiqitang on mice with epithelial ovarian cancer. Methods: Human ovarian cancer cell line SKOV-3 was injected subcutaneously into the right hip of 60 female BALB / C mice to establish epithelial ovarian cancer model. Serum carbohydrate antigen 125 (CA-125), human epididymal epithelial secreted protein 4 (HE4) The proliferating cell nuclear antigen (PCNA), vascular endothelial growth factor (VEGF), matrix metalloproteinase-9 (MMP-9) and the cumulative optical density were detected by SP method. Results: The volume of ovarian cancer mass in CAFQD group [(18.60 ± 0.96) mm3], serum CA125 [(6.13 ± 0.04) U.mL-1] and HE4 [(16.90 ± 0.87) pmol.L- 1], PCNA (12.1 ± 0.56), VEGF (10.00 ± 0.66), MMP-9 (14.10 ± 0.73) and the cumulative optical density in cancer tissue were significantly lower than those in the model group and the treatment group (P <0.01) . Conclusion: QDQT can treat epithelial ovarian cancer by inhibiting cell proliferation and angiogenesis.
