目的:观察山楂饮灌胃联合愈疡生新方灌肠对小鼠免疫性溃疡性结肠炎疾病活动指数、体重变化、结肠长度、结肠炎症及溃疡愈合的作用和量效关系,为临床应用提供实验及理论依据。方法:选择100只昆明雄性小鼠,随机分为5组,即正常组、模型组、山楂饮联合愈疡生新方高剂量组(高剂组)、山楂饮联合愈疡生新方低剂量组(低剂组)、柳氮磺胺吡啶(SASP)组。除正常组外,其余各组均制作免疫性溃疡性结肠炎实验模型。于造模后第2 d开始给药,高剂组小鼠给予山楂饮0.01 g.g-1体重灌胃,愈疡生新方0.008 g.g-1体重灌肠,1次/日;低剂量组小鼠给予山楂饮0.005 g.g-1,愈疡生新方0.004 g.g-1体重灌肠,1次/日;柳氮磺胺吡啶组给予1%SASP灌肠和灌胃各0.002 g.g-1体重,1次/日;正常组、模型组给予生理盐水灌肠和灌胃各0.02 mL.g-1体重,1次/日。连续给药治疗21 d后,观察各组小鼠的疾病活动指数与体重变化。处死小鼠,取全段结肠,称结肠湿重,测肠重指数,观察结肠溃疡及炎症反应情况。结果:小鼠疾病活动指数、体重变化、各组小鼠结肠长度情况、各组小鼠结肠溃疡及炎症反应情况高剂组、低剂组、柳氮磺胺吡啶组明显优于模型组,且高剂组明显高于低剂组、柳氮磺胺吡啶组。结论:山楂饮灌胃联合愈疡生新方灌肠能明显降低小鼠免疫性溃疡性结肠炎的疾病活动指数和溃疡及炎症评分,减轻结肠充血、水肿、增厚等炎性症状,显著改善小鼠免疫性溃疡性结肠炎的结肠缩短程度;具有促进小鼠溃疡愈合、改善炎症反应的作用。其疗效与药量呈正相关。
