将丝网印刷电化学传感器制作技术与戊二醛蒸汽熏蒸的酶固定化方法相结合制作了一次性使用胆碱酯酶电极(AChE),并用于神经性毒剂沙林和梭曼的测定。以氯化硫代乙酰胆碱(ATCh)为底物,该酶电极在pH7.2、温度37℃时有最大响应。酶催化反应的动力学常数为:Km=3.5mmol/L,Imax=2.5μA。沙林、梭曼抑制酶电极响应,百分抑制率与毒剂浓度的对数呈线性关系。当抑制时间为2min时,测定沙林、梭曼的线性范围分别为:5-400μg/L、5~100μg/L。测定湖水样品中的沙林、梭曼的回收率分别为127.4%和125.0%,RSD分别为149%和136%。
A one-time use of AChE was made by a combination of screen printing electrochemical sensor fabrication and glutaraldehyde steam fumigation enzyme immobilization, and was used for the determination of neurotoxic agents sarin and soman. Thiocholine chloride (ATCh) as a substrate, the enzyme electrode at pH7.2, the temperature 37 ℃ when the maximum response. The kinetic constants of enzymatic reaction were: Km = 3.5mmol / L, Imax = 2.5μA. Sarin, soman inhibition of enzyme electrode response, the percentage inhibition rate and the logarithm of the concentration of a linear relationship. When the inhibition time is 2min, the linear ranges of sarin and soman are 5-400μg / L and 5-100μg / L, respectively. The recoveries of sarin and soman in the lake water samples were 127.4% and 125.0%, respectively, with RSDs of 149% and 136%, respectively.
