目的 探讨TGF β1 对肺鳞癌细胞MHCⅠ类抗原表达的影响。方法 以原代培养的人肺鳞癌细胞为对象 ,采用免疫荧光法 ,通过流式细胞仪分别检测加入rhINF γ及rhTGF β1 +rhINF γ肺鳞癌细胞 β2 M的表达状况。结果 rhINF γ能显著诱导肺鳞癌细胞 β2 M的表达 (P <0 0 1) ,但这种诱导作用在各组癌细胞中均能被rhTGF β1 显著抑制 (P <0 0 1) ,且与其分化程度无关。结论 TGF β1 对INF γ诱导人肺鳞癌细胞MHCⅠ类抗原表达有明显抑制作用。
Objective To investigate the effect of TGF β1 on the expression of MHC class Ⅰ antigen in lung squamous cell carcinoma. Methods Human lung squamous cell carcinoma cells were cultured in primary culture. Immunofluorescence staining was used to detect the expression of β2 M in lung squamous cell carcinoma with rhIFFγ and rhTGF β1 + rhIFFγ by flow cytometry. Results rhINFγ significantly induced the expression of β2 M in lung squamous carcinoma cells (P <0.01), but this effect was significantly inhibited by rhTGF β1 (P <0.01) in all cancer cell lines, The degree of differentiation has nothing to do. Conclusion TGF β1 can significantly inhibit the expression of MHC class I antigen in human lung squamous carcinoma cell line INF γ.
