目的:探讨麦冬总黄酮(TFOJ)的提取工艺及其体内外抗氧化作用。方法:采用响应面法优化TFOJ的提取工艺;体外试验测定TFOJ、TFOJE和TFOJB清除自由基的能力;采用D-半乳糖建立小鼠氧化损伤模型,研究TFOJE和TFOJB对其的保护作用。结果:TFOJ的最佳提取条件:提取时间188min,乙醇浓度73%,温度72℃,料液比1∶29,最高得率0.886%。TFOJ、TFOJE和TFOJB均可有效清除DPPH、ABTS+自由基。TFOJB的抗氧化效果较优,可使氧化损伤小鼠血清和肝脏中MDA含量分别下降17.2%、15.7%,GSH-Px、SOD活性分别增高36.5%、17.2%和34.2%、13.5%。结论:麦冬中黄酮类化合物具有良好的自由基清除能力,可显著改善小鼠氧化损伤。
Objective: To investigate the extraction process of Ophiopogon japonicus total flavone (TFOJ) and its antioxidative effect in vitro and in vivo. Methods: The response surface methodology was used to optimize the extraction process of TFOJ. The ability of TFOJ, TFOJE and TFOJJB to scavenge free radicals was measured in vitro. The oxidative damage model of mice was established by D-galactose and the protective effects of TFOJJ and TFOJB were studied. Results: The optimal extraction conditions of TFOJ were: extraction time 188min, ethanol concentration 73%, temperature 72°C, solid-liquid ratio 1:29, and the highest yield 0.886%. Both TFOJ, TFOJE, and TFOJB can effectively eliminate DPPH and ABTS+ radicals. The antioxidant effect of TFOJB was better, and the MDA content in serum and liver of oxidatively injured mice decreased by 17.2% and 15.7%, respectively. The activities of GSH-Px and SOD increased by 36.5%, 17.2%, 34.2%, and 13.5%, respectively. Conclusion: Flavonoids from Ophiopogon japonicus have good free radical scavenging ability and can significantly improve oxidative damage in mice.
