目的:探讨加味丹参饮不同组方对心肌缺血再灌注损伤大鼠环氧酶(COX-2)、细胞间黏附分子-2(ICAM-2)、P38 MAPK表达的影响。方法:根据L_(12)(2~(11))正交设计表格分为12组,加上一个假手术组,将78只白鼠随机分为13组,每组6只,雌雄各3只。采用结扎冠状动脉左前降支的方法制备心肌缺血再灌注模型,观察加味丹参饮不同组方对心肌缺血再冠注损伤大鼠血清中COX-2、ICAM-2、P38 MAPK蛋白的表达。结果:与空白组相比较,加味丹参饮能够显著降低血清中COX-2、ICAM-2、P38 MAPK蛋白的表达(P<0.05),不同组方之间比较,可以得出加味丹参饮中丹参、檀香、川芎、红花四位药物发挥主要作用。结论:加味丹参饮能够显著降低血清中COX-2、ICAM-2、P38 MAPK蛋白的表达而到达治疗作用,在方剂组成中丹参、檀香、川芎、红花发挥主要治疗作用。
Objective: To investigate the effects of different Danshen Yin prescriptions on the expression of cyclooxygenase (COX-2), intercellular adhesion molecule-2 (ICAM-2) and P38 MAPK in rats with myocardial ischemia-reperfusion injury. Methods: According to the L_ (12) (2 ~ (11)) orthogonal design table, we divided into 12 groups, plus a sham operation group, 78 rats were randomly divided into 13 groups, 6 in each group, 3 in each. The model of myocardial ischemia-reperfusion was established by ligating the left anterior descending coronary artery. The expressions of COX-2, ICAM-2 and P38 MAPK in the serum of rats with myocardial ischemia-reperfusion injury were observed. Results: Compared with the blank group, Modified Danshen Decoction could significantly decrease the expression of COX-2, ICAM-2 and P38 MAPK in serum (P <0.05). Compared with the blank group, , Sandalwood, Chuanxiong, saffron four drugs play a major role. Conclusion: Modified Danshen Decoction can significantly reduce the expression of COX-2, ICAM-2 and P38 MAPK in serum and reach the therapeutic effect. Salvia miltiorrhiza, sandalwood, rhizoma chuanxiong and saffron play important roles in the prescription.
