目的观察重组人内抑素(recombinant human endostatin,rh-End)对佐剂性关节炎(adjuvant-induced arthritis,AA)大鼠滑膜组织新生血管生成的抑制作用及其部分机制研究。方法采用弗氏完全佐剂(Freund’s complete adjuvant,CFA)诱导的AA大鼠模型,观察滑膜组织中新生血管生成的病理学改变;免疫组织化学法检测血管内皮细胞生长因子(vascular endothelial growth factor,VEGF)的表达。实时荧光定量PCR对VEGF的表达进行定量检测。结果rh-End能抑制AA大鼠的继发性足肿胀,对滑膜组织中过度生成的新生血管有抑制作用。rh-End能显著减少AA大鼠关节滑膜组织中高表达的VEGF。结论重组人内抑素可显著抑制AA大鼠滑膜组织VEGF的表达及新生血管生成。
Objective To observe the inhibitory effect of recombinant human endostatin (rh-End) on synovial neovascularization in adjuvant-induced arthritis (AA) rats and its partial mechanism. Methods AA rat model induced by Freund’s complete adjuvant (CFA) was used to observe the pathological changes of neovascularization in synovial tissue. The expression of vascular endothelial growth factor VEGF) expression. Real-time fluorescence quantitative PCR for the quantitative detection of VEGF expression. Results rh-End could inhibit the secondary paw swelling in AA rats and inhibit the over-generated neovascularization in synovial tissue. rh-End can significantly reduce the high expression of VEGF in synovial tissue of AA rats. Conclusion Recombinant human endostatin can significantly inhibit the expression of VEGF and angiogenesis in synovial tissue of AA rats.
