研究了聚二甲基硅氧烷(PDMS)膜生物反应器系统在长期封闭循环连续发酵中酵母细胞的适应性进化及发酵动力学。进行了3轮连续发酵实验,每轮发酵时间分别为495、512、440h。采集每轮发酵残液进行菌悬液涂平面培养基的菌株分离与筛选,并转移培养用于下一轮发酵。发酵实验中细胞和乙醇浓度均维持稳定。实验结果表明,筛选出的菌株在生产乙醇的能力上表现出一定优势,3轮发酵单位时间的细胞比产率分别为0.092、0.106、0.126g/g·h。对长期封闭循环连续发酵动力学行为进行了实验分析,获得了一些动力学实验基础参数。
The adaptive evolution and fermentation kinetics of yeast cells in a polydimethylsiloxane (PDMS) membrane bioreactor system over a long period of continuous closed loop fermentation were investigated. Three consecutive fermentation experiments were carried out, with the fermentation time of 495, 512 and 440h respectively. Each round of rape was collected for bacterial suspension coated flat medium strain isolation and screening, and transfer culture for the next round of fermentation. Fermentation experiments in cell and ethanol concentrations were stable. The experimental results showed that the selected strains showed certain advantages in the ability to produce ethanol. The specific cell yields per unit time were 0.092, 0.106 and 0.126 g / g · h, respectively. The kinetic behavior of continuous fermentation under long-term closed cycle was analyzed experimentally, and some basic parameters of kinetic experiment were obtained.
