目的:比较溴化氰活化法和胺还原法制备A群和C群脑膜炎球菌多糖(group A and group C meningococcal polysaccharide,GAMP和GCMP)蛋白结合物的生化及免疫学特性。方法:以GAMP和GCMP为抗原,破伤风类毒素(tetanus toxoid,TT)为载体蛋白,采用溴化氰活化法和胺还原法制备结合物,并对其进行生化及免疫学检测。结果:两种方法均可将多糖与TT有效结合,在小鼠体内均具有良好的免疫原性。溴化氰活化法制备的结合物收率、高分子结合物含量、免疫小鼠产生的抗体效价更高。结论:采用溴化氰活化法制备GAMP-TT,GCMP-TT结合物优于胺还原法。
OBJECTIVE: To compare the biochemical and immunological properties of protein conjugates of group A and group C meningococcal polysaccharide (GAMP and GCMP) between cyanogen bromide activation and amine reduction. Methods: GAMP and GCMP as antigen, tetanus toxoid (TT) as carrier protein, cyanogen bromide activation method and amine reduction method were used to prepare the conjugate and its biochemical and immunological detection. Results: Both methods can effectively combine polysaccharide with TT and have good immunogenicity in mice. The yield of the conjugate prepared by the cyanogen bromide activation method, the amount of the polymer conjugate, and the antibody titer of the immunized mouse are higher. Conclusion: GAMP-TT and GCMP-TT conjugates were better than amine reduction method by cyanogen bromide activation.
