目的:探讨补肾化瘀方对宫腔粘连家兔子宫内膜细胞转化生长因子β1(TGF-β1)及相关Smad蛋白在子宫内膜再生过程中的影响。方法:将10只家兔随机分为空白组、补佳乐组、补肾化瘀方高剂量组、补肾化瘀方中剂量组、补肾化瘀方低剂量组,每组各2只,各组对应连续灌胃7d后取心脏血,离心获取血清。对宫腔镜下取得的宫腔粘连兔子宫内膜组织进行细胞培养并分离间质细胞,分别加入各组血清的培养液培养后采用免疫组化检测TGF-β1、Smad3、Smad7蛋白的表达。结果:补佳乐组、补肾化瘀方高剂量组、补肾化瘀方中剂量组、补肾化瘀方低剂量组TGF-β1、Smad3、Smad7蛋白表达的平均光密度值与空白组比较,差异均有统计学意义(P<0.01);补肾化瘀方低剂量组Smad7蛋白表达,补肾化瘀方中剂量组TGF-β1、Smad3蛋白表达,补肾化瘀方高剂量组TGF-β1、Smad3、Smad7蛋白表达与补佳乐组比较,差异均有统计学意义(P<0.01)。结论:补肾化瘀方可能通过抑制宫腔粘连子宫内膜TGF-β1、Smad3蛋白的表达,上调Smad7蛋白的表达,从而降低子宫内膜细胞纤维化的发生,防止宫腔粘连的形成。
Objective: To investigate the effect of Bushen Huayu Recipe on the process of endometrial regeneration of transforming growth factor β1 (TGF-β1) and related Smad protein in uterine cavity adhesion rabbit endometrium. Methods: Ten rabbits were randomly divided into two groups: blank control group, high-dose group, high-dose Bushen Huayu prescription group, middle-dose Bushen Huayu prescription group and low- Corresponding to 7d after continuous gavage blood, centrifuged to obtain serum. Hysteroscopic intrauterine adhesion of rabbit endometrial tissue obtained by cell culture and stromal cells were isolated, were added to each group of serum culture medium using immunohistochemical detection of TGF-β1, Smad3, Smad7 protein expression. Results: Compared with blank group, the average optical density of TGF-β1, Smad3 and Smad7 protein expression of Bu Jiale group, high-dose Bushen Huayu prescription group, middle-dose Bushen Huayu prescription group and Bushen Huayu prescription low- (P <0.01), the expression of Smad7 in the low dose of Bushen Huayu recipe, the expression of TGF-β1 and Smad3 in the middle dose of Bushen Huayu Recipe, the high dose of Bushen Huayu Recipe in the dose of TGFβ1, Smad3, Smad7 protein expression compared with the nocturnal group, the difference was statistically significant (P <0.01). Conclusion: Bushen Huayu Recipe may inhibit the formation of intrauterine adhesions by inhibiting the expression of TGF-β1 and Smad3 in uterine lining and up-regulating the expression of Smad7 protein.
