近年来麻风树(Jatropha curcas)的生物活性研究如火如荼,但其二萜类化合物的抗癌活性及分子机制研究仍知之甚少.通过萃取和柱层析方法从麻风树的根皮中分离提取到一种二萜类化合物Curcusone C,探讨了该化合物对卵巢癌SK-OV-3细胞增殖、细胞凋亡的影响.实验结果表明,Curcusone C能显著抑制SK-OV-3细胞的增殖,且随着药物浓度的增加其抑制作用呈明显的剂量依赖效应,其48 h半数抑制浓度IC50为160 nmol/L;同时,Curcusone C能显著诱导SK-OV-3细胞凋亡,DAPI染色结合显微镜观察发现400 nmol/L Curcusone C处理后的细胞呈现典型的细胞凋亡形态学特征;最后,Western blot免疫印迹表明Curcusone C能显著诱导SK-OV-3细胞的聚ADP核糖聚合酶(PARP)经Caspase切割为Mr89×103降解产物,从而从分子水平上证实Curcusone C能以剂量依赖方式激活PARP的切割活性进而促进细胞凋亡.上述结果表明从麻风树根皮中提取的二萜类化合物Curcusone C对人卵巢癌的预防与治疗可能具有潜在价值.图5参14
