研究雷公藤内酯醇对T淋巴细胞中核因子 κB活力及其抑制分子IκB的影响 ,进一步阐明雷公藤内酯醇免疫抑制作用的分子机制 .利用凝胶移率实验 (EMSA)检测不同浓度的雷公藤内酯醇对于普通培养状态下和同时使用PMA/PHA激活的Jurkat细胞中NF κB活力的影响 ,并以逆转录 半定量PCR方法检测了雷公藤内酯醇处理后两组Jurkat细胞中IκBα 的mRNA水平的改变 .研究发现 :①在普通培养状态的Jurkat细胞核内存在一定活力的NF κB ,使用PMA/PHA处理可使NF κB活力显著升高 ,雷公藤内酯醇可以降低两种培养状况下的Jurkat细胞中NF κB活力 ,并以激活状态下更为显著 ;②雷公藤内酯醇可以上调Jurkat细胞中IκBαmRNA的表达水平 .雷公藤内酯醇上调IκBα 基因转录有可能是其抑制细胞内NF κB活力的机理之一
To study the effect of triptolide on the activity of NF-κB and its inhibitory molecule IκB in T lymphocytes and further elucidate the molecular mechanism of triptolide’s immunosuppression.METHODS were used to detect the effects of different concentrations of triptolide Alcohol on the activity of NF κB in Jurkat cells activated with PMA / PHA under normal culture conditions and the change of mRNA level of IκBα in two groups of Jurkat cells after triptolide treatment by reverse transcription semi-quantitative PCR The results showed that: (1) NFκB activity in Jurkat cells in normal cultured condition was higher than that in Jurkat cells treated with PMA / PHA, and that triptolide decreased NF kappaB activity and activated more significantly; (2) Triptolide can upregulate the expression of IκBαmRNA in Jurkat cells.It is suggested that triptolide upregulation of IκBαgene may be one of the mechanisms that inhibit the activity of NFκB
