目的 研究流感病毒在裂解剂作用下不同时间的形态变化及对病毒亚单位结构形态上的影响 ,掌握流感病毒在裂解剂作用下的裂解规律。方法 将纯化后的流感病毒抗原按 0 5 %~ 0 6 %比例加入裂解剂TritonN 10 1,分别在 0 5 ,1,2 ,3,3 5 ,4 ,5 ,6h取样 ,用有膜电镜载网粘取样品 ,用 2 %磷钨酸 (PTA)负染 ,电镜下观察。结果 流感病毒裂解可大致分为基质蛋白 (Mprotein ,MP)包膜膨胀阶段 ,神经胺酸酶 (NeuraminidaseNA)、血凝素 (Hemagglu tininHA)、脂质双层 (LipidbilayerLB)脱落阶段 ,基质蛋白包膜破裂核衣壳 (NucleocapsidNP)溢出阶段、基质蛋白溶解阶段。结论 采用震荡措施可使病毒间裂解进程趋于同步。
Objective To study the morphological changes of influenza virus at different times under the action of lysing agents and the morphological changes of virus subunits, and to understand the cleavage regularity of influenza viruses under the action of lysing agents. Methods Purified influenza virus antigens were added into Triton N 10 1 at a ratio of 0% ~ 0 6%, and were sampled at 0, 5, 1, 2, 3, 5, 4, The sample was adhered by a wire, negatively stained with 2% phosphotungstic acid (PTA), and observed under an electron microscope. Results Influenza virus lysis could be divided into three stages: envelope expansion phase of Mprotein (MP), NeuraminidaseNA, Hemagglu tininHA, Lipidbilayer LB phase, matrix protein envelope Ruptured nucleocapsid (Nucleocapsid NP) overflow phase, matrix proteolysis phase. Conclusion The shaking process can make the lysis process between viruses tend to be synchronized.
