目的:观察茴香提取液对糖尿病大鼠早期肝损害的保护作用。方法:采用链脲佐菌素(STZ)诱导大鼠糖尿病模型。将造模成功后的大鼠按血糖值随机分为模型组、二甲双胍组(0.15 g·kg-1)、茴香低剂组(1.5 g·kg-1)和茴香高剂组(3.0 g·kg-1),另取10只为正常对照组。模型组和对照组灌等体积的生理盐水,其他各组按相应药物剂量每天灌胃1次,共14 d。于末次给药2 h后,各鼠采血测空腹血糖值(FBG)、丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、丙二醛(MDA)的含量和超氧化物歧化酶(SOD)活力。取肝脏切片,做HE染色,观察肝脏组织形态学的改变。结果:与正常组比较,模型组显著升高FBG、MDA、AST、ALT水平,(P<0.05,0.01),降低SOD活性(P<0.01),肝组织结构不清,肝细胞溶解坏死明显;与模型组比较,茴香治疗组(高剂组、低剂组)显著增高血清中SOD活性(P<0.01),降低MDA,ALT,AST和FBG水平(P<0.01),明显改善肝组织结构,肝细胞坏死数量少,肝细胞再生明显,高剂量组改善更为明显。结论:茴香对糖尿病大鼠早期肝损害有一定的保护作用。
Objective: To observe the protective effect of fennel extract on early liver damage in diabetic rats. Methods: Diabetic rats were induced by streptozotocin (STZ). The rats after successful modeling were randomly divided into model group, metformin group (0.15 g · kg -1), fennel low dose group (1.5 g · kg -1) and fennel high dose group (3.0 g · kg -1) -1), the other 10 for the normal control group. The model group and the control group were infused with equal volumes of saline, and the other groups were given gavage once daily for 14 days. After 2 h of the last administration, the contents of fasting blood glucose (FBG), alanine aminotransferase (ALT), aspartate aminotransferase (AST), malondialdehyde (MDA) and superoxide dismutase (SOD) activity. Liver slices were taken for HE staining to observe the morphological changes of the liver tissue. Results: Compared with normal group, the levels of FBG, MDA, AST and ALT in model group were significantly increased (P <0.05, 0.01) and SOD activity was decreased (P <0.01), the structure of liver tissue was unclear, Compared with model group, fennel treatment group (high dose group, low dose group) significantly increased serum SOD activity (P <0.01), reduced MDA, ALT, AST and FBG levels (P <0.01), significantly improved liver tissue structure, Small number of liver cell necrosis, hepatocyte regeneration significantly improved in high-dose group more obvious. Conclusion: Fennel on diabetic rats with early liver damage have a protective effect.
