目的 研究急性运动性轴索型神经病(acute motor axonal neuropathy,AMAN)与HLA-Ⅰ类和Ⅱ类等位基因分型的关系,探讨AMAN患者免疫遗传的特点。方法 用改良快速盐析法抽提基因组DNA,采用聚合酶链反应-顺序特异性引物法(PCR-SSP)对33例AMAN患者和132例健康人进行HLA-Ⅰ类和Ⅱ类等位基因分型。结果AMAN组HLA-Ⅰ类基因中B15、B35频率增高,RR值分别为4.09和7.08。Pc(校正P值)分别为0.015和0.00081。AMAN组HLA-Ⅱ类基因中,虽然DR16的频率高于对照组(P=0.004),但校正后的P值失去意义。结论HLA-B15、B35与AMAN的易感性可能有关联。未发现HLA-Ⅱ类基因与AMAN有明显的关联。
Objective To study the relationship between acute motor axonal neuropathy (AMAN) and HLA class Ⅰ and Ⅱ allelic genotypes and to explore the characteristics of immune inheritance in patients with AMAN. Methods Genomic DNA was extracted by a modified rapid salting-out method. The alleles of HLA class I and class II were determined in 33 patients with AMAN and 132 healthy controls by polymerase chain reaction-sequence specific primer (PCR-SSP) type. Results The frequency of HLA-Ⅰ gene B15 and B35 increased in AMAN patients with RR of 4.09 and 7.08, respectively. Pc (corrected P value) is 0.015 and 0.00081, respectively. In AMAN group, although the frequency of DR16 was higher than that of control group (P = 0.004), the corrected P value had no significance. Conclusion HLA-B15, B35 and AMAN susceptibility may be related. No HLA-Ⅱ gene was found to be significantly associated with AMAN.
