目的探讨新生隐球菌荚膜长期保存及染色方法,以方便教学,随时让学生观察到典型新生隐球菌荚膜负染色的形态。方法取新生隐球菌进行小白鼠腹腔接种,吸出腹腔渗出液,用石炭酸固定液杀菌及固定,并用黑色素代替墨汁染色。结果用石炭酸固定液保存的新生隐球菌菌体及荚膜长达20年之久,镜检形态与新鲜标本无异。结论本法可用于教学,黑色素染色法还可用于临床微生物检验。
Objective To investigate the long-term preservation and staining of Cryptococcus neoformans capsule, in order to facilitate teaching, at any time for students to observe the typical Neocortical capsular negative staining morphology. Methods Cryptococcus neoformans was inoculated intraperitoneally in mice, aspirated peritoneal exudate, sterilized and fixed with carbolic acid fixative, and melanin was used instead of ink stain. Results Cryptococcus neoformans cells and capsules preserved with carbolic acid fixation solution for up to 20 years, the microscopic examination of the morphology and fresh specimens is no different. Conclusion This method can be used for teaching, melanin staining method can also be used for clinical microbiological examination.
