目的:观察杞椇颗粒剂对雄性大鼠酒精性肾功能损伤的预防作用。方法:雄性SD大鼠,随机分为杞椇颗粒剂14g/kg、7g/kg、3.5g/kg组,水飞蓟宾胶囊32mg/kg对照组,模型对照组和正常对照组。除正常对照组外,其他各给药组均灌胃56°红星二锅头白酒10ml/kg,同时灌胃相应药物,模型对照组灌胃等体积蒸馏水,连续45天。取肾脏,计算脏器指数,10%甲醛固定后HE染色,光镜下观察肾脏组织病理结构变化;取血清按试剂盒法检测尿素氮(UREA)、肌酐(CR)、尿酸(UA)、葡萄糖(Glu)水平,总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)含量及谷胱甘肽过氧化物酶(GSH-Px)、丙二醛(MDA)、总超氧化物歧化酶(SOD)活力。结果:杞椇颗粒剂14g/kg、7g/kg、3.5g/kg剂量组肾脏指数分别为0.64±0.05、0.65±0.06、0.64±0.09 g/100g,均显著低于模型对照组0.71±0.07 g/100g;MDA活力分别为2.80±2.62、2.78±0.93、2.86±1.25nmol/L,均显著低于模型对照组4.60±3.01 nmol/L;7g/kg剂量组TC、TG含量分别为1.28±0.10、0.38±0.06 mmol/L,均显著低于模型对照组1.54±0.22、0.57±0.30 mmol/L;14g/kg剂量组GSH-Px,SOD活力分别为101.65±34.07μmol/L、155.92±16.98 U/ml,均高于模型对照组80.91±34.80μmol/L、126.21±67.88 U/ml;各剂量组UREA、CR、UA、LDL-C水平均低于模型对照组,Glu、HDL-C水平均高于模型对照组,但差异无统计学意义。光镜下HE法观察肾脏组织病理结构,杞椇颗粒剂各剂量组肾小管有少量上皮细胞出现水样变性及炎性细胞浸润,肾损伤程度较模型对照组明显减轻。结论:杞椇颗粒剂对雄性大鼠酒精性肾功能损伤有明显的预防作用。
Objective: To observe the preventive effect of Qiqi granule on alcoholic renal injury in male rats. Methods: Male Sprague-Dawley rats were randomly divided into 14g / kg, 7g / kg, 3.5g / kg group and 32mg / kg silybin capsule. The model control group and normal control group were randomly divided into four groups. In addition to the normal control group, the other administration groups were fed with 56ml red star Erguotou liquor 10ml / kg, while the corresponding drug was given to the model control group, and the same volume of distilled water was administered for 45 consecutive days. The kidneys were taken out and the organ index was calculated. HE staining was performed after fixed with 10% formalin. The histopathology of renal tissues was observed under light microscope. UREA, CR, UA, (Glu), total cholesterol (TC), triglyceride (TG), high density lipoprotein cholesterol (HDL-C), low density lipoprotein cholesterol (LDL-C) and glutathione peroxidase GSH-Px), malondialdehyde (MDA), total superoxide dismutase (SOD) activity. Results: The kidney index of QIJI granule at doses of 14g / kg, 7g / kg and 3.5g / kg were 0.64 ± 0.05, 0.65 ± 0.06 and 0.64 ± 0.09 g / 100g respectively, which were significantly lower than those of the model control group (0.71 ± 0.07 g / 100g; the MDA activities were 2.80 ± 2.62, 2.78 ± 0.93 and 2.86 ± 1.25nmol / L, respectively, which were significantly lower than 4.60 ± 3.01 nmol / L in model control group. The contents of TC and TG in 7g / kg group were 1.28 ± 0.10 , 0.38 ± 0.06 mmol / L, respectively, which were significantly lower than 1.54 ± 0.22,0.57 ± 0.30 mmol / L in the model control group. The GSH-Px and SOD activities in the 14g / kg group were 101.65 ± 34.07μmol / L and 155.92 ± 16.98U / ml were higher than the model control group 80.91 ± 34.80μmol / L, 126.21 ± 67.88 U / ml; the UREA, CR, UA, LDL-C levels were lower than the model control group, Glu, HDL-C levels Higher than the model control group, but the difference was not statistically significant. Under the light microscope, the pathological changes of kidney tissue were observed by HE method. A small amount of watery degeneration and inflammatory cell infiltration were found in the renal tubular cells of each dose group of QIJI granule. The degree of renal injury was significantly reduced compared with the model control group. Conclusion: Qilu Granule has significant preventive effect on alcoholic renal injury in male rats.
