目的:探讨补充硒以减轻5-氟尿嘧啶(5-FU)对大鼠脂质过氧化损害的机制。方法:60只大鼠随机分为补硒组(Se)和非补硒组(NSe),经腹腔注射5-FU后,分别于注射前及注射后第1、3、5天处死,检测大鼠血液、心、肝、肾及空肠粘膜内丙二醛含量及谷胱甘肽过氧化物酶活性。结果:在Se组,血中及空肠粘膜内丙二醛含量在注射5-FU后第1、3天,心、肝、肾组织内丙二醛含量在注射5-FU后第3天显著低于NSe组;红细胞谷胱甘肽过氧化物酶活性在注射5-FU后一直显著高于NSe组(P<0.01),空肠粘膜在注射5-FU后第1天,心、肝、肾组织内在注射5-FU后第3天,谷胱甘肽过氧化物酶活性显著高于NSe组(P<0.05)。结论:补充硒有助于减轻5-FU对大鼠的脂质过氧化损害。
Objective: To investigate the mechanism of selenium supplementation to reduce the damage of lipid peroxidation induced by 5-fluorouracil (5-FU) in rats. Methods: Sixty rats were randomly divided into selenium group (Se) and non-selenium group (NSe). After intraperitoneal injection of 5-FU, rats were sacrificed on the 1st, 3rd and 5th day after injection, Malondialdehyde content and glutathione peroxidase activity in blood, heart, liver, kidney and jejunum mucosa of rats. Results: In the Se group, the content of malondialdehyde in the blood and jejunal mucosa was significantly lower on the 3rd day after injection of 5-FU In the NSe group, glutathione peroxidase activity of erythrocytes was significantly higher than NSe group (P <0.01) after injection of 5-FU. On the first day after 5-FU injection, On the 3rd day after injection of 5-FU, the activity of glutathione peroxidase in renal tissue was significantly higher than that in NSe group (P <0.05). CONCLUSIONS: Selenium supplementation helps to reduce lipid peroxidation damage induced by 5-FU in rats.
